Der gebürtige Kroate Luka Cicin-Sain studierte an der Universität seiner Heimatstadt Rijeka Medizin und erhielt 1996 seinen Doktortitel. Anschließend studierte er Biomedizin und legte, nach einem Forschungsaufenthalt in Schweden, 2001 sein Staatsexamen in Humaner Medizin am Gesundheitsministerium in Zagreb ab.

Schon während seiner Arbeit als Mediziner faszinierte ihn die Biologie. Er arbeitete bis 2004 am Max von Pettenkofer-Institut für Hygiene und Medizinische Mikrobiologie in München. An der Universität von Rijeka machte er schließlich 2004 seinen Master of Science in Biomedizin, gefolgt von seinem Doktor der Naturwissenschaften im Jahr 2006.

Nach einem fünfjährigen Aufenthalt als Postdoc und wissenschaftlicher Assistenzprofessor am Vaccine and Gene Therapy Institute der Oregon Health and Science University in Portland, USA kam er 2010 als Leiter der Nachwuchsforschergruppe "Immunseneszenz und Chronische Infektionen" ans HZI. Ab Juli 2016 war er Leiter der Gruppe "Immunalterung und Chronische Infektionen" (IMCI). Mittlerweile führt er die Abteilung "Virale Immunologie" (VIRI). Seit 2019 hat Cicin-Sain eine W2-Professur an der Medizinischen Hochschule Hannover (MHH) und ist Mitglied des Zentrums für individualisierte Infektionsmedizin (CiiM), einer gemeinsamen Einrichtung des HZI und der MHH.
