Structure isn’t everything but sure helps
Without doubt Brian Matthews is one the pioneers of structural biology. He has shaped the field from its beginning in the 1960s with numerous seminal contributions towards structure determination, the structural basis of protein stability, protein-DNA interactions and protein engineering. He is the author of the most cited publication in structural biology which describes the so-called "Matthews coefficient", very well known to every protein crystallographer in the world. His studies on T4 lysozyme, using as a model protein to decipher the structural basis of protein stability, are textbook knowledge and paved the way towards rational protein design and protein engineering.
