The department of chemical biology offers a Master-Thesis for students in the field of Organic Chemistry. Title: Organic synthesis of biomimetic carriers for antibiotic conjugates
Description of the project
Infections caused by multidrug-resistant Gram-negative bacteria result in significant mortality and morbidity worldwide despite the development of new anti-infective drugs. The emergence of antibiotic resistance from Gram-negative bacteria introduced one of the biggest challenges in infection research. Therefore, the exploration of innovative strategies for discovering new antibiotics is warranted. This urgent need drove our group to focus on the improvement of small molecule active transport into Gram-negative bacteria. We work on Trojan Horse molecules, consisting of original uptake-mediating structures such as 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid amide core unit, connected to different antibiotics. The uptake-mediating structures act as carriers specifically enforcing the uptake into bacterial cells.
Different bioinspired structures such as from Enterobactin are investigated in our Trojan Horse strategy and new biomimetic carriers have been designed. Such synthetic moieties that are internalized by bacteria will be conjugated via specific linkers to an active agent.
Aim of the thesis
The major goal of the project is to synthesize and fully characterize potential drug carriers that are based on cathecol or hydroxamate motifs, or a combination thereof. The project requires sound practical and theoretical skills in organic chemistry. The student will work synthetically, but will get acquainted to microbiological and imaging characterization techniques.
