Single cell profiling of the immune response to influenza vaccination - proof of concept

The aim of the project is to develop methodology that will allow identifying functional determinants of a nonresponse to influenza vaccination by transcriptomic profiling of peripheral blood mononuclear cells at the single cell level. As a first step, we aim to determine single cell transcriptomes in freshly isolated PBMCs from two healthy volunteers immediately before and 1, 3, 7 days after vaccination. If successful, we will test whether equivalent single cell transcriptomes can be obtained from cryopreserved PBMCs in order to plan the corresponding analyses featuring the biobanked PBMCs from the HZI Flu Study or from cryopreserved PBMCs to be collected in future studies.
